Biological Background: CD19 Function and Localization
- CD19 is a B-lymphocyte antigen and differentiation marker that functions as a coreceptor for the B-cell antigen receptor (BCR) complex.
- It decreases the activation threshold of BCR signaling and activates downstream pathways including phosphatidylinositol 3-kinase and intracellular calcium mobilization. Related references: PMID:12387743, PMID:20101619, PMID:9382888
- CD19 is not essential for early B-cell differentiation in the bone marrow but is required for normal B-1 cell differentiation, peripheral B-cell proliferation, and responses to antigen challenge. Related references: PMID:7542548, PMID:7543183, PMID:9317126, PMID:12387743
- It is required for normal serum immunoglobulin levels and production of high-affinity antibodies. Related references: PMID:12387743, PMID:7542548, PMID:7543183
- CD19 localizes to the cell membrane and membrane rafts.
- It is detected on B cells in spleen, bone marrow, thymus, lymph nodes, and peripheral blood lymphocytes.
- Post-translational modifications include glycosylation and phosphorylation; the protein contains disulfide bonds, immunoglobulin-like domains, and a transmembrane helix.
- The predicted molecular weight is approximately 60 kDa.
Experimental Guidance and Technical Tips
- This APC/Cyanine7-conjugated antibody is suitable for flow cytometry; consider titrating the antibody to determine the optimal concentration for your specific sample.
- Include appropriate isotype controls and viability dyes to ensure accurate gating of live B-cell populations.
- APC/Cyanine7 requires excitation by a 633–635 nm laser and detection with a 780/60 nm or similar filter; ensure your cytometer configuration is compatible.
- The immunogen covers amino acids 19–287 of mouse CD19, targeting the extracellular domain; confirm cell surface expression of CD19 in your model system.
- Validate antibody performance with a known positive control (e.g., mouse splenocytes or B-cell lines) prior to running experimental samples.
