Biological Background: CD38 Function and Localization
- CD38 (ADP-ribosyl cyclase/cyclic ADP-ribose hydrolase 1), also known as ADPRC1, cADPR1, and T10, is a ~34 kDa (also detected at ~13 kDa) transmembrane glycoprotein encoded by the CD38 gene (GeneID: 952).
- It functions as an ectoenzyme with ADP-ribosyl cyclase activity, synthesizing the second messenger cyclic ADP-ribose (cADPR) from NAD+, which plays a critical role in glucose-induced insulin secretion. Related references: PMID:7961800, PMID:8253715
- CD38 also catalyzes the production of the Ca2+-mobilizing messenger nicotinate-adenine dinucleotide phosphate (NAADP+) from NADP+ and nicotinic acid, with a preference for 2'-phospho-cADPR as substrate. Related references: PMID:16690024
- In addition, CD38 exhibits cADPR hydrolase activity, contributing to the regulation of intracellular cADPR levels. Related references: PMID:7961800, PMID:8253715
- Beyond enzymatic functions, CD38 acts as a receptor for CD31 (PECAM-1) on endothelial cells, facilitating lymphocyte adhesion, activation, and transendothelial migration. Related references: PMID:9551996
- In dendritic cells, CD38 signaling modulates mature-stage functions such as CCL21-driven migration, survival, and Th1-polarizing activity. Related references: PMID:16293598
- CD38 is expressed at high levels in pancreas, liver, kidney, brain, testis, ovary, placenta, and certain malignancies including lymphoma and neuroblastoma.
- Subcellularly, CD38 is a cell surface and membrane-localized type II transmembrane protein.
Experimental Guidance and Technical Tips
- The immunogen comprises the extracellular domain (amino acids 43-300), which is expected to be accessible on the surface of live cells, making this antibody suitable for flow cytometry without permeabilization.
- When using the APC/Cyanine7 conjugate, protect the antibody from light and avoid freeze-thaw cycles to maintain tandem dye stability and minimize signal loss.
- For optimal flow cytometry results, titrate the antibody to determine the best concentration for your specific cell type and staining conditions, and include appropriate single-stained compensation controls to account for spectral overlap of APC/Cyanine7.
- Validate reactivity in known CD38-expressing human cell lines (e.g., Jurkat, RPMI-8226, or activated T cells) or primary samples, and confirm specificity using CD38-negative controls.
