Biological Background: AXL Function and Localization

• AXL (also known as UFO, ARK, or JTK11) is a receptor tyrosine kinase encoded by the AXL gene.
• Upon binding its ligand GAS6, AXL undergoes dimerization and autophosphorylation, activating downstream pathways such as PI3K-AKT and recruiting adaptors including GRB2, PLCG1, and LCK.
• AXL signaling regulates diverse processes including cell survival, proliferation, migration, differentiation, and plays a role in innate immune modulation by inhibiting Toll-like receptor-mediated responses.
• Subcellular location: Cell membrane.
• Tissue specificity: Highly expressed in metastatic colon tumors; expressed in primary colon tumors and weakly in normal colon tissue.
• AXL serves as a receptor for various viruses, including Lassa virus, lymphocytic choriomeningitis virus, and Ebolavirus. In Zika virus infection, AXL promotes viral entry into glial cells, Sertoli cells, and astrocytes, and Zika virus potentiates AXL kinase activity to antagonize type I interferon signaling via a SOCS1-dependent mechanism.
