Biological Background: CD54 (ICAM-1) Function and Localization

• ICAM-1 is a cell adhesion molecule that serves as a ligand for the integrin LFA-1 (ITGAL:ITGB2), mediating leukocyte cell-cell adhesion and calibrating T-cell killing strength in response to antigen stimulation.
• It also binds to MAC-1 (ITGAM:ITGB2) on neutrophils, promoting adhesion between stimulated neutrophils and endothelial cells.
• During leukocyte trans-endothelial migration, ICAM-1 engagement triggers assembly of endothelial apical cups via ARHGEF26/SGEF and RHOG activation.
• In epithelial cells, ICAM-1 promotes cell aggregation through interaction with MUC1.
• ICAM-1 is expressed at low levels on a subset of lymphocytes, macrophages, and endothelial cells, and is strongly induced on these cells as well as on fibroblasts and epithelial cells upon stimulation.
