Biological Background: TIM-1/KIM-1/HAVCR Function and Localization
- Hepatitis A virus cellular receptor 1 homolog (HAVCR1), also known as Kidney injury molecule 1 (KIM-1), T-cell immunoglobulin and mucin domain-containing protein 1 (TIMD-1), T-cell immunoglobulin mucin receptor 1 (TIM-1), is encoded by the Havcr1 gene.
- Functions as a phosphatidylserine receptor important in regulatory B-cell homeostasis including generation, expansion, and suppressor functions. Related references: PMID:21821911 PMID:25645598 PMID:32668241
- Involved in efferocytosis by binding to phosphatidylserine on apoptotic cells, promoting their engulfment.
- In injured kidney epithelial cells, confers phagocytic ability to bind and internalize apoptotic bodies and necrotic debris.
- Acts as a P-selectin/SELPLG ligand regulating activated T-cell trafficking during inflammatory responses and T-cell accumulation in the CNS. Related references: PMID:24703780
- Regulates T-cell proliferation and expression of anti-inflammatory cytokines including IL10. Related references: PMID:15793576 PMID:25582854 PMID:25645598
- Localized at cell membrane, phagocytic cup, cytoplasmic vesicle, and phagosome; expressed in stimulated T-cells and constitutively in B-cells. Related references: PMID:11725301 PMID:21821911
- Contains an immunoglobulin domain, disulfide bonds, and glycosylation sites; alternative splicing produces isoforms.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the extracellular domain (amino acids 22-237), making this antibody suitable for detecting cell surface TIM-1 in flow cytometry.
- For optimal staining, titrate the antibody using relevant mouse cell lines or primary cells known to express TIM-1, such as activated T cells or kidney epithelial cells.
- Consider using an Fc receptor blocking step to reduce non-specific binding.
- Note that TIM-1 can be shed from the cell surface under certain conditions; validate staining with appropriate controls.
