Biological Background: Podoplanin Function and Localization
- Podoplanin (Pdpn) is a transmembrane sialomucin-like glycoprotein that functions as a cell surface receptor mediating cell adhesion, migration, and signaling through interactions with multiple partners.
- During development, podoplanin plays a critical role in blood-lymphatic vessel separation by binding CLEC1B on platelets, triggering platelet aggregation. Related references: PMID:14522983 PMID:15231832 PMID:17616532
- In lymph nodes, podoplanin regulates fibroblastic reticular cell contractility and adhesion to the extracellular matrix via CLEC1B engagement and ERM protein modulation. Related references: PMID:25347465
- Podoplanin is required for normal lung cell proliferation and alveolar formation at birth, highlighting its developmental importance. Related references: PMID:12654292
- In keratinocytes, podoplanin promotes an elongated morphology, actin reorganization, increased motility, and decreased adhesion, contributing to epithelial plasticity. Related references: PMID:10574709
- Subcellularly, podoplanin localizes to the plasma membrane, particularly enriched in membrane protrusions such as lamellipodia, filopodia, microvilli, and ruffles, as well as membrane rafts and invadopodia, consistent with its roles in migration and invasion.
- Podoplanin does not function as a water channel or transporter, as it does not affect aquaporin regulation, folic acid, or amino acid transport. Related references: PMID:12032185
- Tissue expression is highest in lung and brain, with lower levels in kidney, stomach, liver, spleen, and esophagus; it is expressed in epithelial cells (choroid plexus, glomerulus, alveolus), mesothelial cells, lymphatic endothelia, and stromal cells of peripheral lymphoid tissue, and is upregulated in colon carcinomas.
Experimental Guidance and Technical Tips
- The immunogen (aa 21-133) corresponds to the extracellular domain of mouse Podoplanin, making this antibody suitable for surface staining in live-cell flow cytometry.
- For flow cytometry, titrate the antibody to determine optimal concentration for your specific cell type. Standard live-cell surface staining protocols are recommended; fixation/permeabilization may be evaluated if intracellular epitopes are targeted.
- This antibody is validated for mouse only; cross-reactivity with other species has not been tested.
- APC conjugation allows direct detection without a secondary antibody, simplifying multicolor panel design.
