- Interleukin-1 receptor type 1 (IL1R1), also known as CD121a, is the primary receptor for the pro-inflammatory cytokines IL1A, IL1B, and the antagonist IL1RN.
- Upon ligand binding, IL1R1 associates with the co-receptor IL1RAP to form a high-affinity signaling complex, triggering NF-kappa-B and MAPK pathways downstream.
- Signaling is mediated through TIR domain interactions, recruiting adaptors TOLLIP, MYD88, IRAK1, or IRAK2, ultimately regulating inflammatory and immune responses.
- The receptor is a single-pass type I membrane protein, also found as a secreted form, and belongs to the immunoglobulin superfamily.
