Biological Background: CD93 Function and Localization

  • CD93, also known as C1q receptor (C1qR), AA4 antigen, or Ly-68, is a ~69 kDa single-pass type I transmembrane glycoprotein belonging to the Group XIV C-type lectin family. Its extracellular region contains multiple EGF-like domains, a C-type lectin domain, and a short cytoplasmic tail.
  • Functions as a receptor for defense collagens, including C1q, mannose-binding lectin (MBL), and surfactant protein A (SFTPA1), thereby mediating phagocytosis of apoptotic cells and immune complexes (PubMed:15004139). Related references: PMID:15004139
  • Plays a central role in endothelial cell biology by promoting angiogenesis and cell migration. It binds multimerin-2 (MMRN2) and IGFBP7, leading to SRC-dependent phosphorylation and CRKL recruitment (By similarity).
  • Regulates endothelial barrier function by modulating VEGFR2 signaling activity (PubMed:38441970). Related references: PMID:38441970
  • Highly expressed in lung, heart, bone marrow, and macrophages; expression is low or absent in brain, liver, thymus, and epithelial cells.
  • Post-translationally modified by N-linked glycosylation and phosphorylation, features that may influence its localization and function (from keywords).
  • Possesses multiple disulfide bonds within EGF-like repeats, conferring structural stability required for ligand interactions.

Experimental Guidance and Technical Tips

  • APC conjugation enables direct detection in flow cytometry (FC) using standard red lasers (633–640 nm). ADC-treated tandem dyes may be used for multicolor panels. Protect the antibody from prolonged light exposure.
  • The immunogen spans the extracellular region (aa 23-181), expected to recognize the CD93 ectodomain. Perform antibody titration (e.g., 0.1–5 µg/mL) to determine the optimal staining concentration for your target cell population.
  • Include appropriate isotype control (rabbit IgG-APC) and viability dye to exclude dead cells and ensure gating accuracy.
  • CD93 is known to undergo ectodomain shedding; therefore, confirm surface expression stability in your system, especially under activation conditions.
