Biological Background: CD86 Function and Localization

  • CD86, also known as B7-2, is a type I transmembrane glycoprotein belonging to the immunoglobulin superfamily.
  • It functions as a costimulatory molecule essential for T-lymphocyte activation, serving as a ligand for the CD28 receptor to provide the secondary signal required for naive T-cell priming (By similarity).
  • CD86 is also involved in regulating B cell function and IgG1 production; upon CD40 engagement, it activates the NF-kappa-B signaling pathway via PLC and PKC (PubMed:23241883). Related references: PMID:23241883
  • Additionally, CD86 acts as an inhibitory ligand for CTLA4, a decoy receptor, thereby blocking CD28-mediated T-cell activation and contributing to immune tolerance (PubMed:7504059, PubMed:7513726). Related references: PMID:7504059 PMID:7513726
  • It is expressed on the cell membrane of antigen-presenting cells, including dendritic cells, macrophages, and B cells.
  • The protein contains an immunoglobulin-like domain, a transmembrane region, and a cytoplasmic tail; it undergoes alternative splicing and post-translational modifications such as glycosylation and ubiquitination.
  • UniProt keywords include adaptive immunity, cell membrane, disulfide bond, glycoprotein, and receptor, reflecting its role in immune signaling.

Experimental Guidance and Technical Tips

  • This APC-conjugated antibody is designed for flow cytometry (FC) applications. Optimal working dilutions should be determined experimentally by the end user.
  • The immunogen corresponds to the extracellular domain (amino acids 24–245) of mouse CD86, ensuring specificity for cell-surface detection.
  • For best results, use fresh or properly cryopreserved cell samples and include appropriate isotype controls and Fc receptor blocking steps.
  • The recommended storage condition is protected from light and kept at 2–8°C for short-term use; avoid repeated freeze-thaw cycles.
