Biological Background: CD59A glycoprotein Function and Localization
- CD59a is a potent inhibitor of the complement membrane attack complex (MAC), protecting self-cells from complement-mediated lysis.
- It binds to C8 components (C8A and C8B) of the assembling MAC and prevents incorporation of multiple C9 copies, blocking formation of the osmolytic pore.
- The protein is anchored to the cell membrane via a GPI anchor and is also secreted.
- It is broadly expressed across many tissues, including liver, kidney, spleen, thymus, brain, and heart, as well as in mononuclear cells, erythrocytes, and platelets, but barely detected in neutrophils.
- CD59a undergoes post-translational modifications including disulfide bond formation, glycosylation, and GPI-anchor attachment; it contains a signal peptide.
- Alternative names include MAC-inhibitory protein (MAC-IP), membrane attack complex inhibition factor (MACIF), and protectin.
