Biological Background: CD47 Function and Localization

  • CD47, also known as integrin-associated protein (IAP), is a multifunctional cell surface receptor that contains an immunoglobulin domain and belongs to the immunoglobulin superfamily.
  • It functions as an adhesive protein mediating cell-to-cell interactions and serves as a receptor for thrombospondin-1 (THBS1), modulating integrin signaling through heterotrimeric G proteins.
  • CD47 plays critical roles in various physiological processes including inflammation, apoptosis, angiogenesis, cardiovascular homeostasis, and immunoregulation. Related references: PMID:20610415 PMID:23591719 PMID:27742621
  • CD47 interaction with SIRPA prevents phagocytosis of red blood cells and inhibits dendritic cell maturation. Related references: PMID:10856220
  • It also interacts with SIRPG to mediate T-cell adhesion and co-stimulation, and positively modulates FAS-dependent apoptosis in T-cells. Related references: PMID:15917238
  • CD47 regulates stem cell self-renewal by inhibiting the expression of pluripotency factors SOX2, OCT4, MYC, and KLF4 in response to THBS1. Related references: PMID:23591719
  • Subcellularly, CD47 is localized at the cell membrane and undergoes alternative splicing, generating multiple isoforms. Post-translational modifications include signal peptide cleavage, disulfide bond formation, glycosylation, and phosphorylation.
  • Additionally, CD47 is involved in nitric oxide-mediated blood pressure regulation and negatively modulates wound healing. Related references: PMID:20610415 PMID:18156939

Experimental Guidance and Technical Tips

  • The immunogen, encompassing the extracellular domain (aa 19-135), is expected to target the exposed region of CD47, making it suitable for detecting cell surface expression in flow cytometry.
  • For flow cytometry, consider using appropriate isotype controls and titrating the antibody to optimize staining conditions.
  • As CD47 is expressed on various hematopoietic and non-hematopoietic cells, validate specificity in the relevant cell type, particularly in blood cells and established cell lines.
