Biological Background: CD44 Function and Localization

  • CD44 is a cell-surface receptor also known as Ly-24, Pgp-1, HERMES, ECMR-III, GP90 lymphocyte homing/adhesion receptor, HUTCH-I, Hermes antigen, and hyaluronate receptor, encoded by the Cd44 gene (Gene ID: 12505) in mouse.
  • It mediates cell-cell interactions, adhesion, and migration by binding extracellular matrix components such as hyaluronan, collagen, growth factors, cytokines, and proteases, and transduces signals via assembled protein complexes. Related references: PMID:25065622 PMID:8343954
  • CD44 participates in lymphocyte activation, recirculation, and homing, as well as hematopoiesis and inflammation, contributing to immune responses and tissue microenvironment sensing.
  • Upon interaction with LGALS9, CD44 activates LCK, ERK, and MAPK signaling pathways to promote natural killer cell activation. Related references: PMID:37006235
  • The protein is localized to the cell membrane, microvilli, and can be secreted, with expression detected in the intestinal epithelium.
  • It undergoes alternative splicing, glycosylation, phosphorylation, sulfation, and proteoglycan modification, and contains disulfide bonds, a signal peptide, and a transmembrane helix.

Experimental Guidance and Technical Tips

  • For flow cytometry, titrate the antibody to determine the optimal dilution for your specific cell type and experimental conditions.
  • Include appropriate isotype controls and viability dyes to ensure accurate gating and minimize non-specific binding.
  • Validate the antibody in your sample system, as CD44 expression levels can vary with cell activation state and tissue type.
