Biological Background: CD4 Function and Localization
- CD4 (T-cell surface glycoprotein CD4), also known as L3T4 or Ly-4, is a type I transmembrane glycoprotein and a member of the immunoglobulin superfamily.
- In T-helper cells, CD4 serves as a coreceptor for MHC class II molecules, binding invariant regions of MHC-II while the T-cell receptor (TCR) recognizes the peptide antigen, thereby enhancing T-cell sensitivity and activation.
- Upon co-engagement with MHC-II, CD4 recruits the Src family kinase LCK to the TCR-CD3 complex, triggering phosphorylation of immunoreceptor tyrosine-based activation motifs (ITAMs) and downstream signaling cascades that promote lymphokine production, T-cell motility, and adhesion.
- CD4 is essential for the positive selection of T-helper cells in the thymus and also contributes to the differentiation of monocytes into mature macrophages.
- Beyond T cells, CD4 is expressed on other immune cells, including macrophages, dendritic cells, and NK cells, where it modulates differentiation, activation, cytokine expression, and cell migration through TCR/LCK-independent mechanisms.
- The protein contains an N-terminal signal peptide, four extracellular immunoglobulin-like domains (D1–D4), a transmembrane helix, and a short cytoplasmic tail with palmitoylation sites; it is N-glycosylated and stabilized by disulfide bonds.
- Alternative splicing generates multiple isoforms, with the canonical isoform having a calculated molecular mass of approximately 51 kDa.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the extracellular domain (amino acids 27–394) of mouse CD4, making this APC-conjugated antibody suitable for surface staining and flow cytometric analysis of live cells.
- For flow cytometry, titrate the antibody to an optimal concentration using the relevant cell population (e.g., mouse splenocytes or thymocytes) and include appropriate isotype controls and viability dyes.
- This antibody is specific for mouse CD4; its reactivity with other species has not been determined. Verify cross-reactivity if needed.
- The APC conjugate is compatible with standard flow cytometry instruments equipped with a red (633 nm) laser; avoid excessive light exposure to prevent photobleaching.
