  • CD4 (T-cell surface glycoprotein CD4), also known as L3T4 or Ly-4, is a type I transmembrane glycoprotein and a member of the immunoglobulin superfamily.
  • In T-helper cells, CD4 serves as a coreceptor for MHC class II molecules, binding invariant regions of MHC-II while the T-cell receptor (TCR) recognizes the peptide antigen, thereby enhancing T-cell sensitivity and activation.
  • Upon co-engagement with MHC-II, CD4 recruits the Src family kinase LCK to the TCR-CD3 complex, triggering phosphorylation of immunoreceptor tyrosine-based activation motifs (ITAMs) and downstream signaling cascades that promote lymphokine production, T-cell motility, and adhesion.
  • CD4 is essential for the positive selection of T-helper cells in the thymus and also contributes to the differentiation of monocytes into mature macrophages.
