Biological Background: CD38 Function and Localization
- CD38 (ADP-ribosyl cyclase/cyclic ADP-ribose hydrolase 1) is a multifunctional transmembrane glycoprotein also known as I-19, ADPRC 1, and NIM-R5 antigen.
- It synthesizes cyclic ADP-ribose (cADPR), a second messenger for glucose-induced insulin secretion, and possesses cADPR hydrolase activity.
- CD38 synthesizes the calcium mobilizer nicotinate-adenine dinucleotide phosphate (NAADP+) from 2'-phospho-cADPR and nicotinic acid, as well as from NADP(+) and nicotinic acid. At both pH 5.0 and 7.4, it preferentially transforms 2'-phospho-cADPR into NAADP+. Related references: PMID:11829748
- As a receptor, CD38 binds CD31 on endothelial cells, promoting lymphocyte activation, proliferation, and migration across the endothelial barrier.
- In lamina propria T lymphocytes, CD38/CD31 interactions initiate signaling via LCK and LAT, leading to cytokine release. It also regulates dendritic cell functions including CCL21-driven migration, survival, and Th1 polarization.
- CD38 is localized to the membrane and features a signal-anchor, a transmembrane helix, disulfide bonds, and glycosylation.
