Biological Background: CD32 Function and Localization
- Low affinity immunoglobulin gamma Fc region receptor II (FcγRIIB), also known as CD32, encoded by the Fcgr2b gene in mouse. Alternative splicing produces multiple isoforms with distinct functional properties.
- Binds the Fc region of complexed immunoglobulins gamma with low affinity, mediating effector and regulatory functions such as phagocytosis of antigen-antibody complexes and modulation of antibody production by B-cells.
- Isoforms IIB1 and IIB1' form caps but fail to mediate endocytosis or phagocytosis, while isoform IIB2 can mediate endocytosis of soluble immune complexes via clathrin-coated pits.
- Coaggregation with B-cell receptors (BCR), T-cell receptors (TCR), or Fc receptors down-regulates activation of B-cells, T-cells, and mast cells, respectively.
- Located at the cell membrane, also found in the cytoplasm, associated with the cytoskeleton, and a secreted isoform (IIB3) is released by macrophages into serum.
- Widely expressed in hematopoietic cells; isoform IIB1 is preferentially expressed in lymphoid lineage, IIB2 in myeloid lineage, and IIB3 is serum-soluble.
- Post-translational modifications include glycosylation, phosphorylation, disulfide bonds, and the presence of a signal peptide and transmembrane domain; belongs to the immunoglobulin superfamily.
