Biological Background: CD22 Function and Localization
- CD22 (Siglec-2, BL-CAM) is a B-cell-specific transmembrane sialoglycoprotein of the immunoglobulin superfamily that serves as an inhibitory coreceptor, highly expressed on B-cells and playing a role in differentiation, antigen presentation, and bone marrow trafficking. Related references: PMID:7533044 PMID:9016707
- It binds α2,6-linked sialic acid residues on surface molecules (e.g., CD22 itself, CD45, IgM) in cis, and can also engage ligands on other cells in trans as an adhesion molecule.
- Phosphorylation of the immunoreceptor tyrosine-based inhibitory motif (ITIM) by LYN kinase recruits protein tyrosine phosphatase SHP-1 (PTPN6), negatively regulating B-cell receptor (BCR) signaling and calcium mobilization.
- Sufficiently strong inhibitory signaling can trigger B-cell apoptosis, highlighting CD22's role in maintaining B-cell tolerance.
- CD22 localizes to the cell membrane and is predominantly expressed on B-lymphocytes.
- The protein undergoes alternative splicing, producing multiple isoforms with observed molecular weights of 89–97 kDa.
- Post-translational modifications include N-glycosylation, disulfide bond formation, and tyrosine phosphorylation, and it contains extracellular immunoglobulin domains.
Experimental Guidance and Technical Tips
- This APC-conjugated antibody is ready for direct flow cytometry analysis of surface CD22 expression on viable mouse B-cells; titrate to determine the optimal concentration for your specific staining protocol.
- For intracellular staining, evaluate fixation and permeabilization conditions as the epitope (amino acids 22–151) may be sensitive to denaturation.
- Consider isotype controls and Fc receptor blocking to minimize non-specific binding in murine samples.
