Biological Background: CD21/CD35 Function and Localization
- Complement receptor type 2 (CR2/CD21/CD35) is a membrane glycoprotein primarily expressed on B-lymphocytes, serving as a receptor for complement C3d, HNRNPU, and IFNA1.
- Upon binding to antigen-associated C3d, CR2 facilitates antigen uptake by B-cells, enhancing B-cell activation and subsequent immune responses.
- It forms the B-cell coreceptor complex with CD19, FCRL5, and CD81, playing a crucial role in B-cell activation and signaling.
- Independently of the BCR, CR2 can activate SRC kinase, which phosphorylates nucleolin/NCL, triggering AKT and GSK3 kinase activities.
- CR2 also functions as a ligand for CD23 (FcepsilonRII), the low-affinity IgE receptor, participating in the regulation of IgE production.
- The protein contains multiple sushi (CCP/SCR) domains, a transmembrane helix, and undergoes disulfide bond formation and glycosylation.
- Encoded by the Cr2 gene in mouse, it is homologous to the human CR2 and is involved in the complement pathway and innate immunity.
Experimental Guidance and Technical Tips
- The immunogen was designed to target the extracellular region (amino acids 21–274), likely covering key sushi domains, making this antibody suitable for detecting surface CD21/CD35 by flow cytometry.
- For flow cytometry, consider titrating the antibody to determine the optimal concentration for mouse B-cell lines or primary splenocytes.
- Since the antibody is conjugated with APC, handle under low-light conditions and use appropriate controls for fluorescence compensation.
- Validate specificity using cells known to express CD21/CD35, such as mouse B-lymphoma lines or primary B-cells, and consider using knockout or blocking controls.
