Biological Background: CD150/SLAM Function and Localization
- CD150/SLAM (signaling lymphocytic activation molecule) is a self-ligand receptor of the SLAM family that modulates activation and differentiation of a wide variety of immune cells, bridging innate and adaptive immune responses.
- In T-lymphocytes, SLAMF1 signaling drives IL-2-independent proliferation and IFN-gamma production, critical for immune activation (PubMed:12351401, PubMed:9126961). Related references: PMID:12351401 PMID:9126961
- The receptor promotes TCR-induced IL-4 secretion in CD4+ T-cells (PubMed:15123745) and is essential for IL-4 production by germinal center T follicular helper cells (PubMed:20525889). Related references: PMID:15123745 PMID:20525889
- In B-cells, CD150/SLAM activates ERK and Akt signaling pathways via SYK and INPP5D, regulating humoral immunity independently of SH2D1A (PubMed:15315965). Related references: PMID:15315965
- CD150/SLAM negatively regulates antigen receptor-mediated IFN-gamma production in double-negative T-cells (PubMed:11477403) and downregulates pro-inflammatory cytokines in LPS-stimulated macrophages (PubMed:15123745). Related references: PMID:11477403 PMID:15123745
- It contributes to innate immunity by recognizing Gram-negative bacteria outer membrane proteins, facilitating phagosome maturation and PI3K complex II recruitment (PubMed:20818396, PubMed:22493499). Related references: PMID:20818396 PMID:22493499
- The type I transmembrane protein is localized at the cell membrane and undergoes glycosylation and phosphorylation, with a mature molecular weight of approximately 36-38 kDa.
Experimental Guidance and Technical Tips
- This antibody is validated for flow cytometry; for optimal staining, titrate the antibody on your target cells and use appropriate isotype controls.
- The antibody recognizes an extracellular epitope (amino acids 25-242), enabling live-cell staining without permeabilization. Keep samples cold and protected from light to preserve APC fluorescence.
- As a monoclonal rabbit antibody with high specificity for mouse CD150, confirm reactivity in your specific mouse strain and cell subset prior to large-scale experiments.
- CD150 is expressed on T cells, B cells, NK cells, macrophages, and dendritic cells; consider co-staining with lineage markers to dissect immune cell populations.
- For intracellular signaling studies, note that the antibody may not be suitable for post-fixation/permeabilization detection; alternative fixation protocols should be tested.
