Biological Background: CD14 Function and Localization
- CD14 is a glycosylphosphatidylinositol (GPI)-anchored glycoprotein also known as myeloid cell-specific leucine-rich glycoprotein.
- Coreceptor for lipopolysaccharide (LPS): CD14 binds monomeric LPS in concert with LBP and delivers it to the LY96/TLR4 complex, initiating innate immune response. Related references: PMID:16148141
- Signal transduction: LPS-bound CD14 triggers signaling via MyD88, TIRAP and TRAF6, leading to NF-kappa-B activation, cytokine secretion and inflammatory response. Related references: PMID:15895089 PMID:8612135
- Lipopeptide coreceptor: CD14 acts as a coreceptor for TLR2:TLR6 heterodimer in response to diacylated lipopeptides and for TLR2:TLR1 heterodimer in response to triacylated lipopeptides, with signaling from the cell surface and subsequent trafficking to Golgi in lipid rafts.
- M. tuberculosis lipoprotein receptor: CD14 serves as an accessory receptor for M.tuberculosis lipoproteins LprA, LprG and LpqH, in conjunction with TLR2 and TLR1, modulating antigen presenting cell functions. Related references: PMID:19362712
- Subcellular localization: CD14 is found on the cell membrane, anchored via GPI, and is also secreted. It localizes to membrane rafts and the Golgi apparatus upon activation.
