Biological Background: CD69 Function and Localization

  • CD69 is a type II transmembrane glycoprotein belonging to the C-type lectin family, encoded by the CD69/CLEC2C gene; it is also known as AIM, EA1, MLR-3, GP32/28, BL-AC/P26, and Early T-cell activation antigen p60.
  • Rapidly induced on the surface of T-cells, platelets, and NK cells upon activation, CD69 stimulates diverse signaling pathways to regulate immune cell homeostasis.
    Related references: PMID:24752896, PMID:26296369, PMID:35930205
  • It modulates T-cell differentiation: CD69 negatively regulates Th17 development through carbohydrate-dependent interaction with galectin‑1 and JAK3/STAT5/RORC signaling, while also promoting conversion of naive CD4 T-cells into regulatory T-cells via the S100A8/S100A9 complex.
    Related references: PMID:24752896, PMID:26296369
  • Acting as an oxidized low-density lipoprotein (oxLDL) receptor in CD4 T‑lymphocytes, CD69 suppresses inflammatory responses by inducing PDCD1 expression through NFAT activation.
    Related references: PMID:35930205
  • CD69 contributes to host defense by directly recognizing P. aeruginosa, leading to ERK1 activation and GM‑CSF secretion; in eosinophils, it induces IL‑10 production through the ERK1/2 pathway.
    Related references: PMID:34841721
  • CD69 negatively regulates chemotactic responses of effector lymphocytes and dendritic cells by acting as a S1PR1 agonist, promoting receptor internalization and degradation.
    Related references: PMID:37039481
  • Localized at the cell membrane, CD69 is expressed on activated T-cells, B-cells, natural killer cells, neutrophils, eosinophils, epidermal Langerhans cells, and platelets.

Experimental Guidance and Technical Tips

  • This APC-conjugated antibody is suitable for direct flow cytometry of surface CD69 on activated human and cynomolgus monkey lymphocytes; consider using appropriate isotype controls and Fc‑blocking reagents.
  • The immunogen spans the extracellular domain (aa 78–199) of human CD69; verify epitope conservation and cross-reactivity in the relevant biological system.
  • For multicolor panels, ensure correct compensation and avoid spectral overlap with the APC channel; confirm staining specificity by comparing stimulated versus unstimulated cell populations.
