Biological Background: CD4 Function and Localization

  • CD4 (T-cell surface glycoprotein, alias T4/Leu-3) is a member of the immunoglobulin superfamily, featuring four extracellular Ig-like domains, a single transmembrane helix, and a cytoplasmic tail that interacts with intracellular signaling molecules.
  • Functions as a coreceptor for MHC class II molecules on antigen-presenting cells: upon antigen recognition, CD4 binds MHC class II and simultaneously associates with the T-cell receptor (TCR), recruiting the Src kinase LCK to phosphorylate CD3 chains and initiate T-cell activation, lymphokine production, and cellular adhesion.
  • Essential for T-helper cell development in the thymus and for the maintenance of peripheral T-helper cell function; also triggers differentiation of monocytes into mature macrophages.
  • Highly expressed on the surface of T-helper cells and, to a lesser extent, on other immune cells including macrophages, dendritic cells, and NK cells, where it modulates differentiation, cytokine expression, and migration.
  • Localized primarily at the cell membrane as a single-pass type I membrane protein; internalization can be triggered upon ligand binding or viral interaction.
  • Key post‑translational modifications include phosphorylation of the cytoplasmic tail, palmitoylation at a membrane-proximal cysteine, and N‑linked glycosylation of the extracellular domains, which are critical for membrane localization and protein interaction.
  • Acts as a receptor for multiple viruses: it is the primary receptor for HIV-1, binding the viral envelope glycoprotein gp120 to mediate viral entry; is down-regulated by HIV-1 Vpu protein; and also serves as a receptor for Human Herpesvirus 7 (HHV-7).   • This antibody is conjugated to Allophycocyanin (APC) and supplied in a format ready for flow cytometry (FC). The immunogen was recombinant human CD4 protein, and the antibody was generated in rabbit as a monoclonal source.
  • For flow cytometry, titrate the antibody to determine the optimal concentration for your specific cell type and experimental conditions. Use an appropriate isotype control conjugate to assess non‑specific binding.
  • The antibody has been validated to cross‑react with human and cynomolgus monkey CD4. For other species, independent validation is recommended.
