Biological Background: CD38 Function and Localization

  • CD38 is a multifunctional transmembrane glycoprotein, also known as ADP-ribosyl cyclase/cyclic ADP-ribose hydrolase 1, with alternative names including 2'-phospho-ADP-ribosyl cyclase and T10.
  • Synthesizes cyclic ADP-ribose (cADPR), a second messenger for glucose-induced insulin secretion, and nicotinate-adenine dinucleotide phosphate (NAADP+), a Ca2+ mobilizer. Related references: PMID:7961800 PMID:8253715 PMID:16690024
  • Functions as a receptor for CD31 on endothelial cells, promoting lymphocyte activation, proliferation, and migration across the endothelial barrier. Related references: PMID:9551996
  • Involved in the regulation of dendritic cell functions, including CCL21-driven migration, survival, and Th1-polarizing activity. Related references: PMID:16293598
  • In lamina propria T lymphocytes, CD38/CD31 cognate interactions activate LCK and LAT, followed by cytokine release. Related references: PMID:11259373
  • Subcellularly, CD38 is localized on the cell surface and cell membrane.
  • Expressed at high levels in pancreas, liver, kidney, brain, testis, ovary, placenta, malignant lymphoma, and neuroblastoma.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the extracellular domain (aa 43–300) of CD38, suggesting suitability for detecting surface CD38 by flow cytometry without permeabilization.
  • For live-cell staining, consider using a viability dye to exclude dead cells and minimize non-specific binding.
  • Titrate the antibody to achieve optimal signal-to-noise ratio in your specific sample type.
  • Cross-reactivity with cynomolgus CD38 enables potential use in non-human primate studies; validate in the relevant model system.
  • As an APC-conjugated antibody, protect from light to maintain fluorophore stability and signal intensity.
