Biological Background: CD14 Function and Localization

• CD14 is a monocyte differentiation antigen, also known as My23 antigen, a myeloid cell-specific leucine-rich glycoprotein anchored to the membrane via a GPI anchor. It belongs to the leucine-rich repeat (LRR) family and is heavily N-glycosylated.
• Serves as a coreceptor for bacterial lipopolysaccharide (LPS), partnering with LBP to deliver LPS to the TLR4/MD-2 complex, leading to MyD88/TIRAP/TRAF6-dependent NF-kB activation and inflammatory cytokine secretion. • Acts as a coreceptor for TLR2:TLR6 heterodimer in response to diacylated lipopeptides and for TLR2:TLR1 heterodimer in response to triacylated lipopeptides, triggering signaling from the cell surface and subsequent targeting to the Golgi in a lipid-raft dependent pathway.
