Biological Background: Siglec-8 Function and Localization
- Siglec-8, also known as SAF-2 or SIGLEC8L, is a member of the sialic acid-binding immunoglobulin-like lectin (Siglec) family.
- It functions as a putative adhesion molecule that mediates sialic-acid-dependent binding to blood cells. Related references: PMID:10625619, PMID:10856141
- Preferentially binds to α-2,3-linked sialic acid and also to α-2,6-linked sialic acid; the binding site may be masked by cis interactions on the same cell surface. Related references: PMID:10625619
- Recognizes glycans with terminal N-acetylneuraminic acid and underlying 6-O-sulfated galactose, with a preference for 6-sulfated sialyl-Lewis X epitopes. Related references: PMID:27357658
- The protein is a type I transmembrane glycoprotein localized to the cell membrane.
- Expressed specifically on the surface of basophils, mast cells, and eosinophils.
- Contains immunoglobulin-like domains, disulfide bonds, and undergoes N-linked glycosylation (keywords: Glycoprotein, Disulfide bond, Repeat, Signal, Transmembrane).
- Alternative splicing generates multiple isoforms (keyword: Alternative splicing).
Experimental Guidance and Technical Tips
- The immunogen covers the extracellular region (aa 17-363), encompassing the sialic acid-binding domains, making it suitable for detecting surface-expressed Siglec-8 in flow cytometry.
- For FC, titrate the antibody to determine optimal working concentration; use appropriate isotype controls and consider cell fixation/permeabilization conditions if needed.
- Reported cross-reactivity is limited to human; validate in other species if required for your experimental system.
