Biological Background: NT5E/CD73 Function and Localization
- Core enzymatic activity: NT5E/CD73 is a 5'-nucleotidase that hydrolyzes extracellular nucleoside monophosphates into nucleosides and inorganic phosphate, with a strong preference for AMP over other substrates. This step is rate-limiting in the generation of immunosuppressive adenosine from ATP.

- Subcellular localization: CD73 is attached to the outer leaflet of the plasma membrane via a C-terminal glycosylphosphatidylinositol (GPI) anchor, positioning its active site in the extracellular space to regulate pericellular nucleotide pools.
- Post-translational modifications: The protein is N-glycosylated and contains structurally important disulfide bonds. Its membrane attachment depends entirely on the GPI anchor.
- Structural features: It belongs to the 5'-nucleotidase family and requires zinc and other divalent metal ions for catalytic activity. The molecule exists as a homodimer on the cell surface.
- Disease relevance: Mutations in NT5E are associated with arterial calcification due to deficiency of CD73 (ACDC), a rare genetic disorder, and the protein is a prominent immune checkpoint target in oncology.
Experimental Guidance and Technical Tips
- The immunogen covers the entire ectodomain (amino acids 27–547), which includes the catalytic and substrate-binding regions. This design favors recognition of native, properly folded CD73 on live cells.
- For flow cytometry, consider titrating the antibody to determine the optimal staining concentration for your cell type of interest. Include appropriate isotype controls and viability dyes.
- CD73 is shed or released on extracellular vesicles in some systems; ensure sample preparation minimizes cell damage to avoid false-negative signals.
- This APC conjugate is well-suited for multiplex panels; verify compensation settings when combining with other fluorophores.
