Biological Background: mGluR2 Function and Localization
- mGluR2 (metabotropic glutamate receptor 2) is a dimeric G protein-coupled receptor that serves as the primary receptor for the excitatory neurotransmitter L-glutamate. Related references: PMID:37286794
- Activation of mGluR2 modulates synaptic transmission and neuronal excitability by inhibiting presynaptic calcium channels and reducing further glutamate release (by similarity).
- The receptor signals via G proteins to down-regulate adenylate cyclase activity, and may be involved in synaptogenesis or synaptic stabilization.
- GRM2, the gene encoding mGluR2, is widely expressed in adult and fetal brain, with protein detection in the brain cortex.
- mGluR2 localizes to the cell membrane and is enriched at synaptic sites, including dendrites and dendritic projections.
- The receptor is post-translationally modified by disulfide bonds and glycosylation, with a predicted molecular weight of approximately 95.6 kDa.
- Notably, mGluR2 has been identified as a host entry factor for several viruses, including influenza virus, rabies virus, and SARS-CoV-2, which hijack its endocytosis for cell entry.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the extracellular domain (amino acids 22–498) of human mGluR2, which may improve specificity for native receptor detection in flow cytometry.
- For flow cytometry, consider optimizing antibody titration and using appropriate isotype controls and cell lines with known mGluR2 expression levels.
- Validate antibody reactivity in the relevant sample system (e.g., human brain tissue or peripheral immune cells) to confirm target recognition.
- Note that the antibody is conjugated to APC; ensure instrument compatibility and use appropriate compensation controls when combining with other fluorophores.
