Biological Background: Integrin-β1 Function and Localization

  • Integrin beta-1 (ITGB1), also known as CD29, fibronectin receptor subunit beta, and VLA-4 subunit beta, is a transmembrane receptor that heterodimerizes with multiple alpha integrin subunits to mediate cell-extracellular matrix adhesion and bidirectional signal transduction.
  • As the common beta subunit, ITGB1 pairs with alpha-1 through alpha-11 and alpha-V integrins to form receptors for collagen, fibronectin, laminin, fibrinogen, vitronectin, and other ligands, thereby regulating cell migration, matrix assembly, and tissue organization.
  • ITGA4:ITGB1 binds to the chemokine fractalkine (CX3CL1) and acts as a coreceptor for CX3CR1-dependent fractalkine signaling, influencing leukocyte trafficking and inflammation. Related references: PMID:23125415 PMID:24789099
  • ITGA5:ITGB1 serves as a receptor for fibrillin-1 and fibronectin, mediating RGD-dependent cell adhesion critical for extracellular matrix integrity and mechanosensing. Related references: PMID:12807887 PMID:17158881 PMID:33962943
  • ITGA5:ITGB1 is also a functional receptor for interleukin-1 beta (IL1B), with binding required for IL1B-mediated signaling, bridging integrin function and innate immunity. Related references: PMID:29030430
  • ITGB1 is targeted by several pathogens: integrin ITGA2:ITGB1 acts as a receptor for human echoviruses, rotavirus, and human parvovirus B19, facilitating viral entry. Additionally, ITGB1 interacts with fungal CotH proteins during mucormycosis. Related references: PMID:32487760
  • Subcellularly, ITGB1 localizes to the plasma membrane, focal adhesions, lamellipodia, and recycling endosomes, and is detected in invadopodia and cleavage furrows, consistent with its roles in adhesion, migration, and cytokinesis.
  • ITGB1 undergoes post-translational modifications including acetylation, phosphorylation, and ubiquitination, which may regulate its stability, trafficking, and signaling functions.
  • Tissue expression: ITGB1 is widely expressed, with reported detection in placenta, skin, liver, muscle, and various cultured cell lines; an isoform specifically expressed in striated muscle (isoform 1D) has been identified. Related references: PMID:33962943 PMID:1551917 PMID:7545396 PMID:7681433

Experimental Guidance and Technical Tips

  • The immunogen spans amino acids 1-728, encompassing nearly the entire extracellular domain of human ITGB1; thus the antibody likely recognizes native conformational epitopes. For flow cytometry, consider using live, non-permeabilized cells to preserve surface antigen integrity.
  • Since the antibody is APC-conjugated, protect from light and avoid freeze-thaw cycles. Titrate the antibody to determine the optimal concentration for your specific cell type and experimental conditions to minimize background.
  • The product is validated for flow cytometry (FC) only; for other applications like Western blot or immunohistochemistry, cross-validate performance in the relevant sample system as reactivity may differ.
  • Confirm species cross-reactivity if working with non-human samples; while stated cross-reactivity is human, sequence homology suggests potential reactivity with other species, but empirical testing is required.
