Biological Background: IL-17A Function and Localization

  • Interleukin-17A (IL-17A), also known as CTLA-8, is a secreted glycoprotein belonging to the IL-17 cytokine family, with a molecular weight of approximately 17.5 kDa (UniProt: Q16552).
  • IL-17A functions as a key effector cytokine of innate and adaptive immunity, critically involved in antimicrobial host defense and maintenance of tissue integrity. Related references: PMID:24120361

Experimental Guidance and Technical Tips

  • The antibody was generated using an immunogen spanning amino acids 24-155 of human IL-17A (mature protein). It is expected to recognize the native secreted form.
  • For intracellular flow cytometry (FC intra), ensure proper fixation and permeabilization steps to allow antibody access to cytoplasmic IL-17A. Consider using standard protocols with paraformaldehyde fixation and saponin or methanol permeabilization.
  • As an APC conjugate, protect the antibody from prolonged exposure to light to prevent photobleaching. Titration experiments are recommended to determine the optimal concentration for your samples.
  • Reported protein weight is 18 kDa; however, glycosylation may result in slightly higher molecular weight bands. Validate specificity in your system using appropriate controls (e.g., stimulated vs. unstimulated T cells).
  • This antibody is validated for human reactivity; cross-reactivity with other species has not been tested. If applied to non-human samples, verify cross-reactivity experimentally.
