- CD95/FAS (TNFRSF6) is a member of the tumor necrosis factor receptor superfamily, also known as Apo-1 antigen or FASLG receptor.
- It functions as the cell-surface receptor for TNFSF6/FASLG, transmitting apoptotic signals upon ligand binding.
- Ligand-induced trimerization recruits the adaptor FADD and procaspase-8, forming the death-inducing signaling complex (DISC), which activates the caspase cascade leading to apoptosis.
- FAS-mediated apoptosis plays a critical role in peripheral immune tolerance and activation-induced cell death of mature T-cells.
- Alternatively spliced secreted isoforms (isoforms 2–6) can act as decoy receptors, blocking apoptosis in vitro.
- The protein localizes primarily to the cell membrane and membrane rafts; soluble isoforms are also secreted.
- Isoform 1 and isoform 6 are expressed at comparable levels in resting peripheral blood mononuclear cells; upon activation, isoform 1 levels increase while isoform 6 decreases.
