  • It functions as the cell-surface receptor for TNFSF6/FASLG, transmitting apoptotic signals upon ligand binding.
  • Ligand-induced trimerization recruits the adaptor FADD and procaspase-8, forming the death-inducing signaling complex (DISC), which activates the caspase cascade leading to apoptosis.
  • FAS-mediated apoptosis plays a critical role in peripheral immune tolerance and activation-induced cell death of mature T-cells.
  • Alternatively spliced secreted isoforms (isoforms 2–6) can act as decoy receptors, blocking apoptosis in vitro.
