Biological Background: CD48 Function and Localization
- CD48 (also known as BLAST-1, SLAMF2, BCM1) is a GPI-anchored cell surface glycoprotein that interacts via its N-terminal immunoglobulin domain with CD244/2B4 and CD2 to regulate immune cell activation and function. Related references: PMID:12007789 PMID:19494291 PMID:27249817
- In T-cells, CD48 associates with CD2 and recruits the Src family kinase LCK and adaptor LAT to the TCR/CD3 complex, promoting LCK phosphorylation and downstream signaling.
- Engagement of CD48 induces phosphorylation of the ITSMs in CD244, triggering formation of the immunological synapse and directed release of cytolytic granules (perforin, granzymes) by T-lymphocytes and NK-cells.
- The protein is primarily localized on the cell membrane and enriched in membrane rafts; a secreted form also exists.
- Widely expressed on all hematopoietic cells, underscoring its role in immune surveillance and intercellular communication.
- Structural features include an immunoglobulin-like domain, disulfide bonds, and a GPI-anchor that mediates membrane attachment. Alternative splicing generates multiple isoforms.
Experimental Guidance and Technical Tips
- The immunogen encompasses the extracellular domain (amino acids 27–220), ensuring recognition of native surface CD48; thus, flow cytometry is the primary recommended application.
- This antibody is directly conjugated to APC, allowing immediate use without secondary reagents. However, if indirect detection is desired, select an appropriate anti-rabbit IgG conjugate.
- For best results, validate the antibody in the relevant human hematopoietic sample type (e.g., PBMCs, lymphoid cell lines) and include proper isotype controls and viability staining.
- Minimize cell manipulation and avoid harsh treatments that may disrupt GPI-anchored protein integrity or membrane rafts.
