Biological Background: CD40 Function and Localization
- Official full name: Tumor necrosis factor receptor superfamily member 5 (CD40); also known as B-cell surface antigen CD40, Bp50, CD40L receptor, CDw40.
- Functions as the receptor for TNFSF5/CD40LG, transducing TRAF6- and MAP3K8-mediated signals that activate ERK in macrophages and B cells, leading to immunoglobulin secretion (By similarity). Related references: PMID:31331973
- Member of the tumor necrosis factor receptor superfamily, characterized by cysteine-rich extracellular repeats and a transmembrane domain.
- Subcellular localization: predominantly a cell membrane single-pass type I membrane protein; a secreted isoform exists due to alternative splicing.
- Tissue specificity: highly expressed on B lymphocytes and detected in primary carcinomas, with broader immune cell expression under activation conditions.
- Keywords and post-translational features: disulfide bond, glycoprotein, alternative splicing, disease variant, signal peptide, reference proteome.
Experimental Guidance and Technical Tips
- The immunogen spans the extracellular domain (amino acids 21-193), suggesting strong recognition of native surface CD40; consider optimal performance in flow cytometry without fixation for live-cell staining.
- For FC, validate the antibody at a range of dilutions (e.g., 0.5–5 µg per 10^6 cells) using positive controls (e.g., human B lymphoblastoid cell lines) and negative controls (e.g., CD40-negative cells).
- If assessing intracellular CD40, include a permeabilization step post-surface staining; note that the immunogen covers the ectodomain but may still detect intracellular isoforms.
- While primarily tested in FC, this monoclonal antibody may cross-perform in other immunoassays (e.g., ELISA, immunohistochemistry) after appropriate optimization; confirm with relevant samples.
