Biological Background: CD270/HVEM Function and Localization
- CD270/HVEM (TNFRSF14) is a member of the tumor necrosis factor receptor superfamily, also known as ATAR, HVEA, LIGHTR, and TR2.
- Functions as a receptor for four distinct ligands: TNFSF14/LIGHT, lymphotoxin-alpha (LTA), BTLA, and CD160, integrating both stimulatory and inhibitory signals in immune regulation. Related references: PMID:10754304 PMID:18193050 PMID:23761635
- Signals via the TRAF2-TRAF3 E3 ligase pathway, promoting immune cell survival and differentiation. Related references: PMID:19915044 PMID:9153189 PMID:9162022
- Acts as a host cell receptor for herpes simplex virus 1 (HSV-1) and HSV-2, facilitating viral entry.
- Localizes to the cell membrane as a single-pass transmembrane receptor.
- Widely expressed, with highest levels in lung, spleen, and thymus; also found on B cells, monocytes, and naive T cells. Related references: PMID:18193050 PMID:19915044
- Undergoes post-translational modifications including glycosylation and phosphorylation; contains disulfide bonds stabilizing its structure.
- In T cells, engagement with LIGHT delivers costimulatory signals, enhancing proliferation and effector functions. On NK cells, interaction with CD160 enhances IFNG production and anti-tumor immunity. Related references: PMID:10754304 PMID:23761635
Experimental Guidance and Technical Tips
- The immunogen spans amino acids 39–162, covering the extracellular ligand-binding domain, which may help preserve recognition of native epitopes on the cell surface.
- For flow cytometry, titrate the antibody to determine the optimal concentration for your specific cell type and experimental conditions to minimize background.
- Use appropriate isotype control (APC-conjugated rabbit IgG) and include viability staining to exclude dead cells.
- CD270 is expressed on various immune subsets; consider co-staining with markers such as CD3, CD19, or CD14 to discriminate T cells, B cells, and monocytes.
- Validate antibody performance in your target sample system (e.g., human PBMCs, splenocytes, or cell lines) before running critical experiments.
