Biological Background: CD228/MFI2 (Melanotransferrin) Function and Localization

• Melanotransferrin (MFI2, CD228, p97) is a cell-surface glycoprotein belonging to the transferrin family, encoded by the MELTF gene. Multiple protein isoforms exist due to alternative splicing, with molecular weights of approximately 80 kDa (full-length) and 32 kDa (soluble form).
• Functions primarily in iron cellular uptake: binds one atom of iron per subunit, internalizes, and recycles to the membrane; may also bind zinc.
• Localizes to the cell membrane, consistent with its role as a membrane-bound iron transporter.
• Undergoes post-translational modifications, including N-glycosylation, GPI-anchor addition (lipoprotein), disulfide bond formation, and phosphorylation.
• Highly expressed in human melanomas and certain fetal tissues; also detected in liver, epithelium, umbilical cord, placenta, and sweat gland ducts.
