Biological Background: IL-13Rα1 Function and Localization

  • Interleukin-13 receptor subunit alpha-1 (IL-13Rα1), also known as cancer/testis antigen 19 (CT19), is a cytokine receptor encoded by the IL13RA1 gene. Alternative splicing may generate multiple isoforms.
  • IL-13Rα1 binds interleukin-13 (IL-13) with low affinity and forms a functional receptor complex with IL4RA, enabling high-affinity IL-13 binding and signaling.
  • The protein also acts as an alternate accessory protein for interleukin-4 (IL-4) signaling, interacting with IL4RA, but cannot substitute for IL2RG in mediating enhanced interleukin-2 (IL-2) binding.
  • IL-13Rα1 is a transmembrane glycoprotein localized at the cell membrane, featuring disulfide bonds and glycosylation that are important for its structure and function.
  • Ubiquitously expressed, IL-13Rα1 shows highest levels in heart, liver, skeletal muscle, and ovary, with lower expression in brain, lung, and kidney; it is also found in B-cells, T-cells, and endothelial cells.
  • As a cancer/testis antigen, IL-13Rα1 is aberrantly expressed in various tumors, making it a potential target for cancer immunotherapy and diagnostic applications.
