Biological Background: IL-13Rα1 Function and Localization
- Interleukin-13 receptor subunit alpha-1 (IL-13Rα1), also known as cancer/testis antigen 19 (CT19), is a cytokine receptor encoded by the IL13RA1 gene. Alternative splicing may generate multiple isoforms.
- IL-13Rα1 binds interleukin-13 (IL-13) with low affinity and forms a functional receptor complex with IL4RA, enabling high-affinity IL-13 binding and signaling.
- The protein also acts as an alternate accessory protein for interleukin-4 (IL-4) signaling, interacting with IL4RA, but cannot substitute for IL2RG in mediating enhanced interleukin-2 (IL-2) binding.
- IL-13Rα1 is a transmembrane glycoprotein localized at the cell membrane, featuring disulfide bonds and glycosylation that are important for its structure and function.
- Ubiquitously expressed, IL-13Rα1 shows highest levels in heart, liver, skeletal muscle, and ovary, with lower expression in brain, lung, and kidney; it is also found in B-cells, T-cells, and endothelial cells.
- As a cancer/testis antigen, IL-13Rα1 is aberrantly expressed in various tumors, making it a potential target for cancer immunotherapy and diagnostic applications.

- The immunogen comprises the extracellular domain (amino acids 29-343), suggesting the antibody recognizes an extracellular epitope, which is ideal for flow cytometry on live, non-permeabilized cells.
- For flow cytometry, use an appropriate isotype control and optimize cell staining conditions (e.g., antibody concentration, incubation time, temperature) using a known positive human cell line.
- Because the antibody is pre-conjugated with APC, it enables direct detection without secondary antibodies, reducing background and simplifying the workflow.
- Validate the antibody in the relevant sample system, considering the ubiquitous but variable expression of IL-13Rα1; human PBMCs, cultured endothelial cells, or tumor cell lines may serve as suitable positive controls.
