Biological Background: CD177 Function and Localization

  • CD177 (also known as NB1 glycoprotein, PRV-1, HNA-2a) is a GPI-anchored glycoprotein expressed on a subset of human neutrophils. It is a neutrophil-specific alloantigen that defines the human neutrophil antigen (HNA)-2 system.
  • CD177 functions as a co-receptor with beta-2 integrin (ITGAM/CD11b and ITGB2/CD18) to mediate TNF-primed neutrophil activation, including degranulation and superoxide production. Related references: PMID:21193407
  • By preventing beta-2 integrin internalization and attenuating chemokine signaling, CD177 promotes neutrophil adhesion over migration. Related references: PMID:28807980
  • CD177 heterophilically interacts with PECAM1 on endothelial cells, contributing to neutrophil transendothelial migration in vitro. Related references: PMID:17580308
  • CD177 serves as a receptor for mature PRTN3 (proteinase 3), displaying it on the neutrophil surface; this interaction may enhance endothelial junctional integrity during diapedesis. Related references: PMID:17244676, PMID:18462208, PMID:23202369
  • Subcellularly, CD177 localizes to the cell membrane, membrane rafts, and secretory cytoplasmic granule membranes; it is also found in lamellipodia. The protein undergoes alternative splicing and is post-translationally modified with a GPI anchor.
  • CD177 is highly expressed in normal bone marrow and weakly in fetal liver. During neutrophil differentiation, its expression begins at the metamyelocyte stage. Notably, CD177 is overexpressed in granulocytes of patients with polycythemia vera (PV) and essential thrombocythemia (ET). Related references: PMID:10753836, PMID:17244676, PMID:12377969
  • Keywords associated with CD177 include innate immunity, cell adhesion, GPI-anchor, alternative splicing, and direct protein sequencing.

Experimental Guidance and Technical Tips

  • The immunogen spans the extracellular domain (aa 22–407) of human CD177, making this APC-conjugated antibody suitable for detecting surface CD177 by flow cytometry.
  • Consider using concentrations of 0.5–5 µg per million cells, but optimal dilution should be experimentally determined.
  • Verify antibody reactivity in your specific experimental system, particularly when analyzing CD177 expression across different neutrophil subpopulations.
  • For intracellular staining or Western blotting, performance of this APC conjugate has not been characterized; alternative antibody formats may be more appropriate.
  • Note that CD177 is expressed on a variable percentage of neutrophils in healthy individuals; include appropriate controls.
