Biological Background: SIRPα/CD172a Function and Localization

  • Tyrosine-protein phosphatase non-receptor type substrate 1 (SIRPα, also known as SHPS-1, CD172a, BIT, MYD-1, p84) is an immunoglobulin-like cell surface receptor that acts as a docking protein for PTPN6 and PTPN11, inducing their translocation from cytosol to plasma membrane.
  • Functions as a receptor for CD47, mediating negative regulation of phagocytosis, mast cell activation, and dendritic cell activation, and inhibiting maturation and cytokine production by dendritic cells upon CD47 binding.
  • Supports cerebellar neuron adhesion, neurite outgrowth, and glial cell attachment, suggesting roles in synaptogenesis and synaptic function.
  • Acts as a receptor for thrombospondin-1 (THBS1), which stimulates SIRPA phosphorylation and triggers NADPH oxidase-derived ROS production in non-phagocytic cells. Related references: PMID:24511121
  • Exists as three main isoforms (Sirp-alpha-1, -2, -3) due to alternative splicing, with ubiquitous tissue expression but highest levels in brain and on myeloid cells, and absent on T-cells.
  • Localizes to the cell membrane as a single-pass type I membrane protein, with an extracellular region containing three immunoglobulin-like domains and a cytoplasmic tail with tyrosine phosphorylation sites.

Experimental Guidance and Technical Tips

  • The immunogen covers amino acids 31–370, which comprises the entire extracellular domain; therefore, the antibody is expected to recognize the native conformation of SIRPα on the cell surface.
  • As an APC-conjugated monoclonal antibody, it is ready for direct immunofluorescence staining in flow cytometry without the need for a secondary detection reagent.
  • For optimal staining, titrate the antibody on the target cell type (e.g., human monocytes, macrophages, or dendritic cell lines) to determine the working concentration.
  • Include an appropriate isotype control (APC-conjugated rabbit IgG matched clone) and, if using whole blood or tissue-derived cell suspensions, pre-incubate with Fc receptor blocking reagent to minimize non-specific binding.
  • Since SIRPα is not expressed on T-cells, they can serve as an internal negative control; validate specificity under your experimental conditions using known positive and negative cell populations.
