Biological Background: CD117/c-Kit Function and Localization

• CD117/c-Kit, also known as Mast/stem cell growth factor receptor Kit, is a transmembrane receptor tyrosine kinase encoded by the KIT proto-oncogene. It belongs to the type III receptor tyrosine kinase family and is characterized by extracellular immunoglobulin-like domains, a single transmembrane helix, and a split intracellular kinase domain.
• The primary function of CD117/c-Kit is to serve as the cell-surface receptor for stem cell factor (SCF/KITLG). Upon ligand binding, KIT dimerizes, leading to autophosphorylation and activation of multiple downstream signaling cascades, including PI3K/AKT, RAS/MAPK, and JAK/STAT pathways.
• Through these pathways, CD117/c-Kit regulates essential cellular processes such as cell survival, proliferation, differentiation, and migration. It plays critical roles in hematopoiesis, stem cell maintenance, gametogenesis, melanogenesis, and mast cell development and function.
