Biological Background: Annexin A1 Function and Localization

  • Annexin A1 (also known as Annexin I, Lipocortin I, Calpactin II) is a member of the annexin family of Ca2+-dependent phospholipid-binding proteins.
  • It mediates glucocorticoid anti-inflammatory effects and has anti-inflammatory activity, contributing to the down-regulation of the early phase of the inflammatory response. Related references: PMID:8425544
  • In the adaptive immune response, Annexin A1 enhances signaling cascades triggered by T-cell activation, promotes Th1 cell differentiation, and negatively regulates Th2 differentiation. Related references: PMID:17008549
  • Annexin A1 functions by activating formyl peptide receptors, promoting chemotaxis, actin cytoskeleton rearrangement, cell polarization, and migration; it also contributes to inflammation resolution and wound healing, and enhances CXCL2 release in neutrophils. Related references: PMID:15187149, PMID:22879591, PMID:25664854
  • It negatively regulates hormone exocytosis through activation of formyl peptide receptors and reorganization of the actin cytoskeleton. Related references: PMID:19625660
  • Annexin A1 has high affinity for Ca(2+), binding up to eight Ca(2+) ions, and displays Ca(2+)-dependent binding to phospholipid membranes. Related references: PMID:2532504, PMID:8557678
  • It plays a role in the formation of phagocytic cups and phagosomes, mediating Ca(2+)-dependent interaction between phagosomes and the actin cytoskeleton.
  • Annexin A1 is found in the nucleus, cytoplasm, cell projections (including cilia and phagocytic cups), various membrane domains, endosome membrane, cytoplasmic vesicle membrane, and is secreted into the extracellular space and exosomes.
  • Detected in resting neutrophils, peripheral blood T cells, extracellular vesicles in serum from inflammatory bowel disease patients, placenta, and liver. Related references: PMID:10772777, PMID:17008549, PMID:25664854, PMID:2532504

Experimental Guidance and Technical Tips

  • The immunogen encompasses amino acids 2–346 of human Annexin A1, covering the majority of the protein, which may support recognition of native epitopes in flow cytometry applications.
  • Consider using human peripheral blood leukocytes or cell lines expressing Annexin A1 as positive controls for FC assay optimization.
  • As Annexin A1 can localize to the cell surface and intracellular compartments, both surface and intracellular staining protocols may be considered, with appropriate fixation and permeabilization if targeting intracellular pools.
  • Given the APC conjugation, plan for single-stain controls and compensation adjustments in multicolor panels to avoid spectral overlap.
