Biological Background: P-selectin Function and Localization
- P-selectin (CD62P, also known as GMP-140, LECAM3, PADGEM) is a single-pass type I membrane protein encoded by the SELP gene. It belongs to the selectin family of adhesion molecules.
- Functions as a Ca²⁺-dependent receptor that binds to carbohydrate ligands (sialyl-Lewis X) on neutrophils and monocytes, facilitating leukocyte rolling and the initial steps of inflammation.
- Mediates the interaction between activated endothelial cells or platelets and leukocytes through its counter-receptor SELPLG (P-selectin glycoprotein ligand-1).
- Also interacts with integrins alpha-IIb/beta3 (ITGA2B:ITGB3) and alpha-V/beta-3 (ITGAV:ITGB3) to promote stable cell adhesion. Related references: PMID:37184585
- Subcellular location: Predominantly stored in the alpha-granules of platelets and Weibel-Palade bodies of endothelial cells. Upon cellular activation by agonists, it is rapidly translocated to the cell membrane.
- Contains structural features including a lectin domain, an EGF-like domain, multiple sushi (SCR) repeats, and a transmembrane helix. Post-translational modifications include disulfide bond formation, glycosylation, and palmitoylation.
