Biological Background: CD45RO Function and Localization

• CD45RO is an isoform of receptor-type tyrosine-protein phosphatase C (PTPRC, CD45), generated by alternative splicing. It is also known as leukocyte common antigen (L-CA) or T200.
• PTPRC/CD45RO is a protein tyrosine phosphatase critical for T-cell activation. It positively regulates coactivation via DPP4 binding, and the first phosphatase domain dephosphorylates key signaling proteins including SKAP1, FYN, and LYN, while the second domain modulates substrate specificity.

• CD45RO localizes to the cell membrane, and is enriched in membrane rafts and at the immunological synapse, positioning it to mediate signal transduction during T-cell responses.
• Acts as a receptor for human cytomegalovirus (HCMV) protein UL11, which upon binding reduces tyrosine phosphorylation of signaling proteins and impairs T-cell proliferation.
• The protein is post-translationally modified by N-glycosylation and phosphorylation, features essential for its stability and interactions. UniProt keywords include glycoprotein, phosphoprotein, and host-virus interaction.
