Biological Background: CD11c (Integrin alpha-X) Function and Localization
- The protein is integrin alpha-X, also known as CD11c, Leu M5, and leukocyte adhesion glycoprotein p150,95 alpha chain. It is encoded by the ITGAX gene and belongs to the integrin alpha chain family.
- Integrin alpha-X/beta-2 functions as a receptor for fibrinogen by recognizing the G-P-R sequence, mediating cell-cell interactions during inflammatory responses, and playing a critical role in monocyte adhesion and chemotaxis.
- It also acts as a receptor for the erythrocyte-specific adhesion molecule ICAM4, mediating erythrophagocytosis. Related references: PMID:16985175
- The protein is predominantly expressed in monocytes and granulocytes, consistent with its role in leukocyte adhesion and immune responses.
- Subcellularly, integrin alpha-X is localized at the cell membrane, where it forms heterodimers with integrin beta-2.
- Key structural features include calcium and magnesium binding sites, disulfide bonds, and a transmembrane helix, classifying it as a metal-binding glycoprotein and cell adhesion receptor.
Experimental Guidance and Technical Tips
- The immunogen is a recombinant full-length human CD11c protein; consider validating antibody specificity in the relevant cell types (e.g., monocytes, dendritic cells) using knockout or blocking controls.
- For flow cytometry (FC), optimize the antibody titration and use appropriate isotype controls to ensure specific staining. APC conjugation enables direct detection.
- Although recommended for FC, the antibody may potentially work in other applications; however, validation in the intended assay system is required.
