Biological Background: CD11b Function and Localization
- CD11b, also known as integrin alpha-M (ITGAM), is a member of the integrin family that pairs with the beta-2 subunit (ITGB2/CD18) to form complement receptor 3 (CR3), also designated MAC-1.
- ITGAM/ITGB2 mediates adhesive interactions of monocytes, macrophages, and granulocytes and facilitates uptake of complement-coated particles and pathogens (PubMed:20008295, PubMed:9558116). Related references: PMID:20008295 PMID:9558116
- CD11b acts as a receptor for iC3b fragment of complement component 3, recognizing the R-G-D peptide, as well as fibrinogen and factor X, thereby linking innate immunity and coagulation.
- The protein regulates neutrophil migration (PubMed:28807980) and, together with ITGB2, is required for CD177-PRTN3-mediated activation of TNF-primed neutrophils (PubMed:21193407). Related references: PMID:28807980 PMID:21193407
- ITGAM/ITGB2 serves as a receptor for ICAM1, promoting adhesion between stimulated neutrophils and endothelial cells (PubMed:1980124). Related references: PMID:1980124
- CD11b is located on the cell membrane and enriched in membrane rafts, consistent with its role in signal transduction and cell adhesion.
- Predominantly expressed in monocytes and granulocytes (PubMed:1346576) and detected in neutrophils at the protein level (PubMed:21193407). Related references: PMID:1346576 PMID:21193407
- Belongs to the integrin alpha chain family, containing calcium and magnesium binding sites, disulfide bonds, and glycosylation modifications; associated with systemic lupus erythematosus.
Experimental Guidance and Technical Tips
- The immunogen is recombinant human CD11b protein; consider that the antibody may recognize both native and denatured forms, but native conformation is critical for flow cytometry.
- For flow cytometry (FC), titrate the antibody to determine optimal staining concentration using the relevant cell population, such as isolated monocytes or granulocytes.
- Include appropriate isotype controls and viability dyes to ensure specific staining and accurate gating in FC experiments.
- CD11b expression can be upregulated upon activation; plan experiments considering dynamic expression levels.
- Cross-reactivity has been confirmed only for human; if working with other species, validate specificity independently.
- Shipping on ice bag ensures antibody stability during transit; upon receipt, store at recommended conditions (typically 2-8°C or aliquot and freeze).
