Biological Background: CD30 Function and Localization
- CD30 (Tumor necrosis factor receptor superfamily member 8, also known as Ki-1 antigen or CD30L receptor) is a transmembrane receptor encoded by the TNFRSF8 gene.
- It functions as a receptor for TNFSF8/CD30L (PubMed:8391931), and ligand binding may play a role in regulating cellular growth and transformation of activated lymphoblasts. Related references: PMID:8391931
- CD30 signaling activates NF-kappa-B, thereby influencing gene expression in immune cells (PubMed:8999898). Related references: PMID:8999898
- Subcellularly, CD30 is localized to the cell membrane and cytoplasm.
- At the protein level, CD30 is detected in alveolar macrophages, indicating tissue-specific expression.
- The CD30 protein is subject to extensive post-translational modification, including glycosylation, disulfide bond formation, and phosphorylation. Alternative splicing and alternative initiation produce multiple isoforms, with molecular weights observed around 14 kDa, 50 kDa, and 63 kDa.
- Structural features include an N-terminal signal peptide, multiple cysteine-rich repeats, a transmembrane domain, and a cytoplasmic tail.
Experimental Guidance and Technical Tips
- This APC-conjugated rabbit monoclonal antibody is designed for flow cytometry (FC). Validate optimal antibody concentration by titration on CD30-positive cell lines or stimulated lymphocytes.
- As CD30 is expressed on activated T and B cells, consider using PMA/ionomycin stimulation or Hodgkin lymphoma-derived cell lines (e.g., L428, KM-H2) as positive controls.
- Include appropriate isotype control (APC-conjugated rabbit IgG) and Fc receptor blocking reagent to minimize non-specific binding.
- For whole blood or PBMC staining, a red blood cell lysis step may be performed after surface staining, provided the fluorophore stability is confirmed.
