Biological Background: Siglec-7/CD328 Function and Localization
- Siglec-7 (Sialic acid-binding Ig-like lectin 7), also known as CD328, AIRM-1, p75, or D-siglec, is a type I transmembrane protein belonging to the immunoglobulin superfamily and siglec family of sialic acid-binding lectins.
- Functions as an inhibitory receptor on immune cells, preferentially binding α-2,3- and α-2,6-linked sialic acids on target cells; ligand engagement triggers tyrosine phosphorylation of its ITIM motifs, recruiting SH2-domain-containing phosphatases to block activating signals.
- Plays a key role in regulating natural killer (NK) cell cytotoxicity, inhibiting killing of sialic acid-expressing targets; also suppresses differentiation of CD34+ hematopoietic precursors toward the myelomonocytic lineage and inhibits proliferation of leukemic myeloid cells.
- Expressed predominantly on resting and activated NK cells, and at lower levels on granulocytes and monocytes; high expression observed in placenta, liver, lung, spleen, and peripheral blood leukocytes.
- Localized to the plasma membrane as a single-pass transmembrane protein; extracellular region contains Ig-like C2-type domains responsible for sialic acid recognition, which may be masked by cis interactions on the same cell surface.
- Post-translational modifications include N-linked glycosylation, disulfide bond formation, and phosphorylation, with the phosphotyrosine sites serving as docking sites for SH2-domain proteins.
- Alternative splicing generates multiple transcript variants, potentially modulating ligand-binding properties or expression levels.
- Structural studies (3D-structure available) reveal the molecular basis for sialic acid specificity, aiding in understanding its immunomodulatory functions.
Experimental Guidance and Technical Tips
- This APC-conjugated antibody (clone not specified) was raised against the extracellular domain (aa 18-144) of human Siglec-7, making it suitable for detecting cell surface expression by flow cytometry.
- For FC, titrate the antibody to determine optimal concentration; use a matched isotype control to set gates and assess background.
- Validate staining in known positive cell lines or primary human NK cells; cross-reactivity is reported for human only, so ensure the experimental system involves human samples.
- Consider that Siglec-7 may undergo cis-interactions masking epitopes; pretreatment with sialidase may enhance detection in some cell types.
- This product is provided as an APC conjugate; handle with care to avoid photobleaching, and use appropriate compensation controls in multicolor panels.
