Biological Background: LAG3 Function and Localization

  • Encoded by the LAG3 gene, lymphocyte activation gene 3 protein (also known as CD223) is a type I transmembrane protein of the immunoglobulin superfamily with a molecular weight of approximately 57 kDa.
  • Acts as an inhibitory receptor on antigen-activated T-cells, delivering negative signals upon binding to its main ligand MHC class II on antigen-presenting cells or FGL1, thereby inhibiting T-cell receptor (TCR) signaling and T-cell activation.
