Biological Background: LAG3 Function and Localization
- Encoded by the LAG3 gene, lymphocyte activation gene 3 protein (also known as CD223) is a type I transmembrane protein of the immunoglobulin superfamily with a molecular weight of approximately 57 kDa.
- Acts as an inhibitory receptor on antigen-activated T-cells, delivering negative signals upon binding to its main ligand MHC class II on antigen-presenting cells or FGL1, thereby inhibiting T-cell receptor (TCR) signaling and T-cell activation. - Ligand binding induces ubiquitination of the KIEELE motif, exposing the RRFSALE motif and promoting formation of condensates with the TCR component CD3E, which disrupts the CD3E-LCK association and blocks TCR activation. Related references: PMID:40101708, PMID:40592325
- Negatively regulates the proliferation, activation, effector function, and homeostasis of both CD4+ and CD8+ T-cells, and contributes to the suppressive function of regulatory T-cells (Tregs) as well as regulation of plasmacytoid dendritic cells.
