Biological Background: CD19 Function and Localization
- B-lymphocyte antigen CD19, also known as Differentiation antigen CD19, is a cell surface protein specifically expressed on B cells.
- Functions as a coreceptor for the B-cell antigen receptor complex (BCR) on B-lymphocytes, lowering the threshold for activation of downstream signaling pathways and triggering B-cell responses to antigens.
- Activates signaling pathways that lead to phosphatidylinositol 3-kinase activation and the mobilization of intracellular Ca(2+) stores. Related references: PMID:12387743, PMID:20101619, PMID:9382888
- Not required for early steps during B cell differentiation in bone marrow, but essential for normal differentiation of B-1 cells. Related references: PMID:7542548, PMID:7543183, PMID:9317126
- Required for normal B cell differentiation and proliferation in response to antigen challenges. Related references: PMID:12387743, PMID:7542548, PMID:9317126
- Required for normal levels of serum immunoglobulins and for production of high-affinity antibodies in response to antigen challenge. Related references: PMID:12387743, PMID:7542548, PMID:7543183
- Subcellular location: Localizes to the cell membrane and is enriched in membrane rafts.
- Tissue specificity: Detected on B cells in spleen, bone marrow, thymus, and lymph nodes, and on peripheral blood lymphocytes (at protein level). Related references: PMID:12387743, PMID:20101619, PMID:7542548, PMID:7543183
- Belongs to the immunoglobulin superfamily, possessing multiple immunoglobulin-like domains, disulfide bonds, and phosphorylation sites (glycoprotein, phosphoprotein).
Experimental Guidance and Technical Tips
- The immunogen corresponds to the extracellular domain (amino acids 19–287), making the antibody suitable for detecting surface-expressed CD19 on live B cells.
- Directly conjugated to APC, it is ready for flow cytometry without a secondary antibody; consider titrating to optimize signal-to-noise in your specific cell population.
- Validate performance in relevant mouse B-cell samples (splenocytes, bone marrow, lymph node cells, peripheral blood) with appropriate isotype and compensation controls.
- For multicolor panels, ensure minimal spectral overlap with other fluorophores and include fluorescence-minus-one (FMO) controls for accurate gating.
- Since CD19 expression persists throughout B-cell development, it can serve as a pan-B-cell marker across differentiation stages.
