Biological Background: CD79a (B-cell antigen receptor alpha chain, MB1) Function and Localization

  • CD79a (also known as Ig-alpha, MB-1, or IGAlpha) is the alpha chain of the B-cell antigen receptor complex (BCR), essential for antigen recognition and signal initiation in B-cells.
  • In cooperation with CD79B, CD79a triggers the signal transduction cascade upon antigen binding, leading to BCR internalization, trafficking to late endosomes, and antigen presentation. It is required for surface expression of the BCR and for efficient differentiation of pro- and pre-B-cells.
  • CD79a stimulates SYK autophosphorylation and activation, and binds BLNK to bring it into proximity with SYK, enabling BLNK phosphorylation. It also interacts with and increases activity of some Src-family tyrosine kinases.
  • During immature B-cell development, CD79a can repress BCR signaling, indicating a regulatory role in early B-cell maturation.
  • Localized to the cell membrane, CD79a is a single-pass type I transmembrane protein with an extracellular immunoglobulin-like domain and an immunoreceptor tyrosine-based activation motif (ITAM) in its cytoplasmic tail.
  • Expressed specifically in B-cells, rendering it a pan B-cell marker widely used in diagnostic and research settings.
  • The protein undergoes several post-translational modifications including glycosylation, phosphorylation, ubiquitination, and methylation, which can influence its function and electrophoretic mobility.
  • The precursor protein has a predicted molecular weight of ~25 kDa; mature glycosylated forms may appear as a band around 25 kDa or a doublet at 20–25 kDa in SDS-PAGE.

  • This antibody was raised against a recombinant fusion protein corresponding to the intracellular C-terminal region (aa 165–225). It is designed for intracellular staining by flow cytometry (FC intra). Consider permeabilization with saponin or methanol for intracellular detection.
  • For detection of surface CD79a on live cells, an antibody targeting the extracellular domain may be more appropriate.
  • Being a rabbit monoclonal antibody, it offers high specificity and lot-to-lot consistency.
  • Validate the antibody in a relevant human B-cell line (e.g., Raji, Daudi) or primary peripheral blood B-cells under the intended experimental conditions.
  • No cross-reactivity with other species has been reported; therefore, its suitability for non-human samples is untested.
  • The antibody is supplied as an APC conjugate, enabling direct detection without secondary antibodies. Protect from light and minimize freeze-thaw cycles.
