Biological Background: CD24 Function and Localization
- CD24, also known as signal transducer CD24 or small cell lung carcinoma cluster 4 antigen, is a GPI-anchored glycoprotein encoded by the CD24 gene (aliases: CD24A).
- May have a pivotal role in cell differentiation of different cell types, with signaling triggered by lectin-like ligand binding to CD24 carbohydrates and transduced by GPI-anchor-derived second messengers.
- Modulates B-cell activation responses: promotes antigen-dependent proliferation of B-cells and prevents their terminal differentiation into antibody-forming cells. Related references: PMID:11313396
- In association with SIGLEC10, may be involved in the selective suppression of the immune response to danger-associated molecular patterns (DAMPs) such as HMGB1, HSP70, and HSP90, and plays a role in the control of autoimmunity (By similarity).
- Localized to the cell membrane via a GPI-anchor.
- Expressed on B-cells, a number of B-cell lines (P32/ISH, Namalwa), T-cells, erythroleukemia cell lines, and small cell lung carcinoma cell lines.
- Computed molecular weight of the unprocessed precursor is approximately 8 kDa; glycosylated mature forms migrate around 13 kDa.
- Features alternative splicing, and is a glycoprotein, lipoprotein, and GPI-anchored membrane protein.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal extracellular domain (amino acids 1–80) of human CD24, which contains the mucin-like domain; this region is likely accessible for flow cytometry detection on live cells.
- For flow cytometry applications, consider titrating the antibody to determine the optimal concentration for your specific cell type, and include appropriate isotype controls.
- CD24 expression is predominantly on B-cells and certain cancer cell lines; validate antibody performance in your relevant sample system.
- This product ships on ice bags; upon receipt, store according to the manufacturer’s recommendations to maintain stability.
