Biological Background: ADAM10 (CD156c) Function and Localization
- ADAM10 (Disintegrin and metalloproteinase domain-containing protein 10), also known as CD156c, CDw156, Kuzbanian, and MADM, is a transmembrane metalloprotease belonging to the ADAM family. It plays critical roles in ectodomain shedding of numerous substrates.
- Acts as a sheddase for adhesion proteins, growth factor precursors, and cytokines, including TNF-α, heparin-binding EGF-like growth factor, ephrin-A2, CD44, and N-cadherin (PubMed:11786905, PubMed:12475894, PubMed:20592283). Related references: PMID:11786905, PMID:12475894, PMID:20592283
- Functions as the major α-secretase for amyloid precursor protein (APP), thus regulating the production of neurotoxic Aβ peptides and linking it to Alzheimer's disease pathology (PubMed:11786905, PubMed:26686862, PubMed:29224781). Related references: PMID:11786905, PMID:26686862, PMID:29224781
- Involved in the proteolytic processing of Notch, mediating lateral inhibition during neurogenesis and required for marginal zone B cell development (By similarity).
- Cleaves FasL, generating the transmembrane remnant FasL APL (PubMed:17557115). Also processes other substrates including JAM3, cellular prion protein, CORIN, ITM2B, IL6R, and IL11RA (PubMed:20592283, PubMed:11477090, PubMed:19114711, PubMed:21288900, PubMed:26876177). Related references: PMID:17557115, PMID:20592283, PMID:11477090, PMID:19114711, PMID:21288900, PMID:26876177
- Localizes to the cell membrane, Golgi apparatus, clathrin-coated vesicles, adherens junctions, and axon/dendrite projections, indicating roles in both intracellular trafficking and cell surface proteolysis (PubMed:28600292). Related references: PMID:28600292
- Expressed in spleen, lymph node, thymus, peripheral blood leukocytes, bone marrow, cartilage, and fetal liver, with detectable protein in brain (PubMed:23676497, PubMed:11511685, PubMed:9016778). Related references: PMID:23676497, PMID:11511685, PMID:9016778
- Promotes the cytotoxic activity of S.aureus alpha-hemolysin by binding the toxin at zonula adherens and facilitating pore formation (By similarity).
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 214–672 of human ADAM10, encompassing the metalloprotease and disintegrin domains. This is predicted to generate antibodies recognizing the extracellular region of the protein, suitable for detecting cell surface ADAM10 by flow cytometry.
- As a directly conjugated APC antibody, it is ready for use in flow cytometry without secondary detection steps. Include appropriate isotype controls and perform titration to determine optimal working concentration.
- ADAM10 is broadly expressed in hematopoietic cells and solid tissues; thus, validate antibody specificity in relevant control cell lines or primary cells expressing varying levels of the target.
- For analysis of ADAM10 shedding activity, consider combination with specific inhibitors (e.g., GI254023X) to confirm signal modulation.
