- CD101, also known as immunoglobulin superfamily member 2 (IGSF2) or cell surface glycoprotein V7 (EWI-101), is a single-pass type I membrane protein belonging to the immunoglobulin superfamily.
- It functions as a negative regulator of T-cell activation, inhibiting CD3-induced proliferation, IL2RA expression, and IL2 secretion through the suppression of tyrosine kinases and phospholipase C-gamma-1 phosphorylation.
- CD101 prevents the nuclear translocation of NFAT, thereby blocking downstream calcium signaling and cytokine production in T-cells.
- Additionally, CD101 contributes to the inhibition of T-cell proliferation via IL10 secretion by cutaneous dendritic cells, indicating a role in peripheral tolerance.

- Since CD101 is expressed on activated T-cells but not on resting T-cells, consider using in vitro–activated human peripheral blood mononuclear cells (PBMCs) or cell lines such as Jurkat (with appropriate stimulation) as positive controls.
- The provided APC conjugate allows direct detection without a secondary antibody; typical starting titrations for flow cytometry range from 0.5 to 5 µg/mL, but optimal concentration must be determined empirically.
- For intracellular staining or analysis of CD101 cleavage products, additional validation may be required, as this antibody targets the extracellular region.
